Mark-recapture techniques are used for studies of animal populations. With only three sampling occasions, both Bailey's triple-catch (BTC) and Jolly-Seber's (J-S) stochastic method can be applied. As marking and handling of fragile organisms may harm them, and thereby affect their chances of being recaptured, handling should be minimized. This can be achieved by taking a subsample before the main sample at the second sampling occasion. Individuals in the main sample are marked and released, whereas those in the subsample are only used for identifying recaptures. Monte-Carlo simulation was used to compare the subsampling method with the ordinary mark-recapture methods. Model-generated populations were sampled with and without subsampling to provide estimates of population size, loss, and dilution rates. The estimated parameters were compared with their true values to identify biases associated with the sampling methods, using 81 different combinations of population size, dilution rate, loss rate, and sampling effort. Each combination was replicated 1,000 times. In no cases did subsampling perform more poorly than the ordinary methods. J-S was slightly more accurate than BTC to estimate the population size, but only when sampling effort was high. The relative biases associated with estimates of dilution and loss rates were substantial, but declined with increasing population size and sampling effort. Confidence limits for the population parameters generally were reliable and tended to be conservative. We therefore conclude that ordinary mark-recapture methods can be supplemented with subsampling without sacrificing accuracy. Subsampling is especially advantageous in cases where marks are difficult to observe under field conditions.  相似文献

Development and reproductive traits of Tetranychus macfarlanei Baker & Pritchard (Acari: Tetranychidae) were investigated on kidney bean, Phaseolus vulgaris L., at eleven constant temperatures. Tetranychus macfarlanei was able to develop and complete its life cycle at temperatures ranging from 17.5 to 37.5°C. At 15 and 40°C, a few eggs (2–4%) hatched but further development was arrested. Development from egg to adult was slowest at 17.5°C and fastest at 35°C for both females and males. Using Ikemoto and Takai’s linear model, the estimated lower developmental thresholds for egg-to-female adult, egg-to-male adult and egg-to-egg development were 12.9–13.0°C. The thermal constants for the respective stages were 110.85, 115.99 and 125.32 degree-days (DD). The intrinsic optimum temperatures (T Φ) calculated by non-linear SSI model were determined as 24.4, 24.4 and 24.2°C for egg-to-female adult, egg-to-male adult and egg-to-egg development, respectively. The net reproductive rate (R 0) was highest at 25°C (167.4 females per female) and lowest at 17.5°C (42.6 females per female). The intrinsic rate of natural increase, r m, increased linearly with the rising of temperature from 0.102 at 17.5°C to 0.441 day−1 at 35°C. These values suggested that T. macfarlanei could be growing quickly in response to increasing temperatures from 17.5 to 35°C and provide a basis for predicting its potential geographical range.  相似文献

Recently diverged taxa may continue to exchange genes. A number of models of speciation with gene flow propose that the frequency of gene exchange will be lower in genomic regions of low recombination and that these regions will therefore be more differentiated. However, several population-genetic models that focus on selection at linked sites also predict greater differentiation in regions of low recombination simply as a result of faster sorting of ancestral alleles even in the absence of gene flow. Moreover, identifying the actual amount of gene flow from patterns of genetic variation is tricky, because both ancestral polymorphism and migration lead to shared variation between recently diverged taxa. New analytic methods have been developed to help distinguish ancestral polymorphism from migration. Along with a growing number of datasets of multi-locus DNA sequence variation, these methods have spawned a renewed interest in speciation models with gene flow. Here, we review both speciation and population-genetic models that make explicit predictions about how the rate of recombination influences patterns of genetic variation within and between species. We then compare those predictions with empirical data of DNA sequence variation in rabbits and mice. We find strong support for the prediction that genomic regions experiencing low levels of recombination are more differentiated. In most cases, reduced gene flow appears to contribute to the pattern, although disentangling the relative contribution of reduced gene flow and selection at linked sites remains a challenge. We suggest fruitful areas of research that might help distinguish between different models.  相似文献

Estimates of the proportion of amino acid substitutions that have been fixed by selection (α) vary widely among taxa, ranging from zero in humans to over 50% in Drosophila. This wide range may reflect differences in the efficacy of selection due to differences in the effective population size (N(e)). However, most comparisons have been made among distantly related organisms that differ not only in N(e) but also in many other aspects of their biology. Here, we estimate α in three closely related lineages of house mice that have a similar ecology but differ widely in N(e): Mus musculus musculus (N(e) ~ 25,000-120,000), M. m. domesticus (N(e) ~ 58,000-200,000), and M. m. castaneus (N(e) ~ 200,000-733,000). Mice were genotyped using a high-density single nucleotide polymorphism array, and the proportions of replacement and silent mutations within subspecies were compared with those fixed between each subspecies and an outgroup, Mus spretus. There was significant evidence of positive selection in M. m. castaneus, the lineage with the largest N(e), with α estimated to be approximately 40%. In contrast, estimates of α for M. m. domesticus (α = 13%) and for M. m. musculus (α = 12 %) were much smaller. Interestingly, the higher estimate of α for M. m. castaneus appears to reflect not only more adaptive fixations but also more effective purifying selection. These results support the hypothesis that differences in N(e) contribute to differences among species in the efficacy of selection.  相似文献

The first reported examples of C-terminal aldehyde analogs of an insect neuropeptide are described. They are hexapeptide insect kinin analogs Boc-VFFPWG-H and Fmoc-RFFPWG-H. Activity observed for these modified analogs in an in vitro insect diuretic assay confirms that the C-terminal aldehyde group is tolerated by an insect kinin receptor. The two analogs demonstrate greatly enhanced activity over standard C-terminal amide insect kinins in a larval weight gain inhibition assay in the corn earworm Helicoverpa zea. Treatment with Boc-VFFPWG-H led to significant increases in larval mortality at doses of 500pm (45%) and 5nm (67%). Boc-VFFPWG-H represents a lead analog in the development of novel, environmentally friendly pest insect management agents based on the insect kinin class of neuropeptides.  相似文献

Identifying the genes responsible for adaptation has been an elusive goal in evolutionary biology. Rock pocket mice (Chaetodipus intermedius) provide a useful system for studying the genetics of adaptation: most C. intermedius are light-coloured and live on light-coloured rocks, but in several different geographical regions, C. intermedius are melanic and live on dark-coloured basalt lava, presumably as an adaptation for crypsis. Previous work demonstrated that mutations at the melanocortin-1 receptor gene (Mc1r) are responsible for the dark/light difference in mice from one population in Arizona. Here, we investigate whether melanism has evolved independently in populations of dark C. intermedius from New Mexico, and whether the same or different genes underlie the dark phenotype in mice from these populations compared with the dark mice from Arizona. Seventy-six mice were collected from pairs of dark and light localities representing four different lava flows and adjacent light-coloured rocks; lava flows were separated by 70-750 km. Spectrophotometric analysis of mouse pelage and of rock samples revealed a strong positive association between coat colour and substrate colour. No significant differences were observed in the colour of rocks among the four lava flows, suggesting that mice in these separate populations have experienced similar selection for crypsis. Despite this similarity in environment, melanic mice from the three New Mexico populations were slightly, but significantly, darker than melanic mice from Arizona. The entire Mc1r gene was sequenced in all mice. The previously identified mutations responsible for the light/dark difference in mice from Arizona were absent in all melanic mice from three different populations in New Mexico. Five new Mc1r polymorphisms were observed among mice from New Mexico, but none showed any association with coat colour. These results indicate that adaptive melanism has arisen at least twice in C. intermedius and that these similar phenotypic changes have a different genetic basis.  相似文献

Feeding by spider mites can cause severe injury to a host plant and lead to a decreasing per capita growth rate and an increasing per capita emigration rate. Such density-dependent responses to local conditions are important in a metapopulation context because they allow the herbivores to colonize new host plants and thereby prolong the time until regional (metapopulation) extinction. In order to include density-dependent responses of the two-spotted spider mite (Tetranychus urticae) in a realistic metapopulation model, a series of greenhouse experiments was conducted with the purpose to quantify how the condition of bean plants (Phaseolus vulgaris) influences the demographic parameters of T. urticae. Plant age per se reduced the growth rate of the spider mites only slightly, whereas the growth rate declined significantly as the plants were injured by the mites. The relationships between plant condition (expressed by the plant injury index D) and the birth and loss (death + emigration) rates of the mites were quantified so as to predict population growth as a function of D. Maximum per capita growth rate (r) was estimated to be c. 0.21 day−1. The growth rate is expected to be negative when D exceeds 0.8. When mites were allowed to emigrate to neighbouring plants via bridges, the per capita emigration rate increased almost exponentially with D. The proportion of eggs in the population decreased with D while the numerical ratio between immatures to adults and the sex ratio did not change with D. Overall, immatures and adults constituted 74% and 26%, respectively, of the active mites and c. 46% of the adults were males. The bridges that connected a donor plant with the surrounding recipient plants were responsible for the majority of the emigrations from donor plants. Most mites stopped after having crossed a single bridge, but a few crossed two bridges while none crossed three bridges within 24 h. Short-term effects on consumption and oviposition rates of four strainsof Neoseiulus californicus (McGregor) after a diet shiftwere evaluated. The new feeding conditions experienced by the predators weresixfixed densities of eggs or protonymphs of Tetranychusurticae Koch placed on excised strawberry leaflet discs andmaintained under laboratory conditions (25±1°C,75–85% RH, 16L: 8D). The observations were made on the first and thefifthday of the experiment. The phytoseiids came from three long-term mass-rearedstrains fed on T. urticae, Dermatophagoides farinaeHughes,or Quercus spp. pollen, respectively. The fourth strainwascollected directly in a strawberry field. Time since diet transfer can be addedto the factors (i.e. feeding history and prey density) already known to affectthe functional and numerical responses of N. californicus,both when it feeds on prey eggs and protonymphs. If consumption rates wereaveraged over all strains and densities, 9.04 and 11.41 eggs, and 6.97 and 6.48protonymphs were consumed on the first and the fifth day, respectively. If thesame was done for oviposition rates, predators feeding on eggs produced 1.46and2.36 eggs/female/day, whereas predators feeding on protonymphs produced 1.35and2.29 eggs/female/day. Time had the greatest impact on the functional responseofthe strain that had previously fed on tetranychids, while an effect of time onthe numerical response was detectable in all strains.  相似文献

We have previously demonstrated that plasminogen activator inhibitor (PAI-1) is associated with the extracellular matrix of cultured bovine smooth muscle cells (Knudsen, B.S., Harpel, P.C., Nachman, R.L. (1987) J. Clin. Invest. 80, 1082-1089). In this report we describe the physiologic role of PAI-1 during the interaction of the tissue plasminogen activator (t-PA) secreting Bowes human melanoma cell line with endothelial extracellular matrices. In addition we have characterized the t-PA.PAI complexes formed during this interaction in the presence and absence of plasminogen. In the absence of plasminogen, a 104-kDa complex between Bowes t-PA and PAI-1 appears in the supernatant. In the presence of plasminogen, PAI initially prevents plasmin formation on the matrix and protects the matrix from degradation by plasmin. The 104-kDa t-PA.PAI complex is degraded into a 68 and a 47-kDa complex by small amounts of plasmin generated from secreted Bowes t-PA and plasminogen. Analysis of these complexes revealed that t-PA is rapidly cleaved by plasmin within the complex whereas complexed PAI-1 is not further degraded. Matrix-associated PAI-1 may play an important role in the protection of extracellular matrices from remodeling and degradation by cellular t-PA and plasminogen.  相似文献
